Cell Line Studies

Cell Line Studies has been the methodology which incorporates tools used in cellular and molecular biology, providing excellent model systems for studying the normal physiology and biochemistry of cells. The cell line studies are also used for drug screening and development and large scale manufacturing of biological compounds such as vaccines, therapeutic proteins.

Course Goal
The course focuses upon:
  • The aspirant gets a hold in aseptic techniques, cell isolation, propagation and maintenance in culture.
  • Apart from biological fundamentals of cell cultivation, the course lays emphasis on the communication control points of laboratory organization
  • Conducting of experiments on the reproducible cultivation of various cell types
Course Curriculum
  • Maintenance of cell lines, testing cell viability
  • Determination of IC50 for different drugs or compounds in different cell lines
  • Cell survival assays
  • Total cell lysate preparation
  • Measurement of drug concentration
  • Gene and protein expression studies processes
Module: 2 Laboratory
  • Recombinant protein production in bench-top fermenters
  • Fermenter setup and sterilization
  • Fed-batch strategy and experimental design
  • Inoculum development, fermenter inoculation, pH and DO preparation, and calibration
  • Continuation of bench scale fermentation-harvest
  • Oxygen mass transfer in 10-liter fermentor
  • Pilot scale (100L) fermentors
  • Fermentor setup, sterilization and preparation for inoculation
  • Fermentation monitoring and control
  • Troubleshooting issues
  • Harvest and processing of 100-liter fermenters
  • Continuous flow centrifugation
